Palonosetron Hydrochloride Overdose Reaction

There is no known antidote for palonosetron, so supportive therapy should be used for overdose. In the Palonosetron dose exploration study, 50 adult cancer patients were given a dose of 90 μg/kg (equivalent to a fixed dose of 6 mg), which was close to 25 times the recommended dose of 0.25 mg. The adverse events of this dose group were similar to those of other dose groups, and there was no dose dependence. Although no research on dialysis treatment has been conducted, dialysis cannot be used as an effective treatment for drug overdose due to the large volume of distribution of palonosetron. The lethal dose of a single dose of intravenous palonosetron for rats and mice is 30 mg/kg (calculated by body surface area, the doses for rats and mice are equivalent to 947 and 474 times the recommended dose for humans), and the main symptoms of poisoning For convulsions, wheezing, pale complexion, cyanosis, and collapse.
